Title:
C: 157 - Genotyping
Description:
Summarize statistics for fish genotyped to date, give progress assessment. Approximately 900 new fish genotyped by end of each reporting quarter.
WE Agreement Type:
Contracted
Deliverable Specification:
Genotype 3600 new fish. Includes the most recent year's samples plus fish from the backlog of samples from the 1990's. In particular, we will concentrate on finishing the last of the Hold summer run fish from the early 1990’s. A fish has been successfully genotyped following initial PCR of 8 microsatellite loci, running of products on an ABI 3100 capillary electrophoresis system, initial scoring of electrophoretic output, re-running and scoring for any failed reactions or ambiguous peaks, input of data into the master database, and error checking of the database. More details on loci and methods are explained in Araki et al., (2006; Conservation Biology, in press; http://oregonstate.edu/~blouinm/pdf_files/Araki%20et%20al%20In%20press%20Cons%20Biol.pdf ).
